dc.description.abstractThe research work was carried out to determine the genetic variability of 20 selected Spondias dulcis L. germplasm collected from the south-western region of Bangladesh during October 2011 to March 2012. Hog plum is a perennial fruit tree in subtropical and tropical areas. In Bangladesh, it has been used as edible fruits. The classification and functional identification of hog plum remains unsolved these days. In the present study, morphological and molecular characterizations were used to record different levels of variability concerning the morphological traits of the fruit. Random Amplified Polymorphic DNA (RAPD) marker was used for the molecular identification. Among the 20 selected germplasm, fourteen were commercially cultivated germplasm. The 10-mer and 12-mer oligonucleotide primers were used in RAPD for amplification. Three primers, A01, A02, and S01, were able to direct the amplification and yielded a total of 362 band patterns of which 63.06% were polymorphic. The highest percent of polymorphic loci (66.67%) was observed for primer A02 and the lowest (60.00%) was from primer S01. Results were analyzed by molecular algorithm UPGMA and Neighbor-Joining. Twenty genotypes on the dendrogram were identified and divided into two major groups and subgroups on the basis of genetic characteristics. The range of genetic distance was observed 0.091 (GP09 vs. GP04) to 0.389 (GP14 vs. GP13). Based on the cluster analysis, the S. dulcis samples that were believed to have high morphological difference were grouped independently. The results suggested that RAPD is useful for the discrimination of cultivars and selection of potential hog plum clones of high economy for crop improvement.
