Study of the Persistency of an Environmental Isolate of Vibrio Cholerae under Different Biotic and Abiotic Conditions

Abstract

Vibrio cholerae was identified in water, sediment and water hyacinth samples from different locations of Moyuri River. Among them V. cholerae from water hyacinth was detected throughout various seasons. A V. cholerae isolate from surface water was used for further studies to determine its persistency under different biotic and abiotic conditions. The persistence of V. cholerae at different detergent, pesticides concentrations and in association (direct and indirect) with Anabaena spp. was investigated. The growth of V. cholerae was not found to be affected in the presence of pesticides both in LB and MS media during the first 4 hours of incubation. However, 24 hour of treatment showed slight reduction in growth. The growth of V. cholerae was found to be reduced at a higher detergent concentration (40%) compared to lower concentrations (10%, 20%, and 30%). The growth in the presence of detergent was further reduced over time with initial 6× 104 c. f. u on Day 1 to 2x104 c. f. u on Day 4 at 40% detergent concentration. The growth of isolated V. cholerae was greatly enhanced in direct association with two and three week's old culture of Anabaena sp. PCC 7120. It can be anticipated that biotic and abiotic conditions may have some effect on growth of V. cholerae.
