Isolation of Potential Probiotic (Clostridium butyricum) for Controlling Pathogenic Bacterial Growth Using PCR Technique in Prawn Farm

Abstract

The present study was carried out from January to November, 2014 to evaluate the antagonistic effect of probiotic species, Clostridium butyricum, on vibrio spp. by means of in-vitro and in-vivo test as well as to determine the impact of this probiotic species on the growth and immunity of fresh water prawn (Macrobrachium rosenbergii) based on enzyme activity and haemocyte count respectively. Both the pathogenic and probiotic bacterial species were isolated from fresh water prawn collected from local market of Khulna. The probiotic incorporated basal diet and diet without any probiotic served as treatment group and control group respectively. These two types of diets were fed to M. rosenbergii separately under treatment group and control group for a period of about 60 days. After the feeding trail, the antagonistic effect as well as the growth parameters such as weight gain, specific growth rate and enzyme activity (amylase and protease) were found to be significantly (P <0.05) higher in treatment group compared with control group. Although immunity determining parameter (haemocyte count) was exhibited to be non-significantly (P> 0.05) higher in treatment from control. Ultimately this study revealed that probiotic, C. butyricum incorporated diets were beneficial for M. rosenbergii culture in terms of decreasing pathogenic bacterial load, increasing growth, enzyme activity and immunity. Thus the present study will be helpful to improve fresh water prawn farming.
