Abstract

Achras sapota is a perennial fruit tree in subtropical and tropical areas. In Bangladesh, it has been used as edible fruits. The classification and functional identification of Achras sapota remains unsolved these days. In the present study, genetic variation and relationship among 15 sapota germplasm were analyzed using Random Amplified Polymorphic DNA (RAPD). Three oligonucleotide primers A02, A03 and S07 were used to direct the amplification which yielded a total of 57 band patterns out of that 48.89% were polymorphic. The highest percent of polymorphic bands (66.67%) was observed for primer A03 and the lowest (20%) was from primer S07. Results were analyzed by molecular algorithm UPGMA. Fifteen genotypes on the dendrogram were identified and divided into two major groups and subgroups on the basis of morphological characteristics. The range of genetic distance was observed 0.112 to 0.5. Based on the cluster analysis, the Achras sapota samples that were believed to have high morphological difference were grouped independently. The results suggested that RAPD is useful for the discrimination of uncultivated, cultivars and potential Achras sapota of high economy.
